چکیده

The objective of this study was to develop solid self-emulsifying pellets to deliver the poorly bioavailable Milk Thistle extract (Silybum Marianum) [1]. These pellets were prepared via extrusion/spheronization procedure, using a self-emulsifying system or SES (composed of Akoline MCM, Miglyol, Tween 80, Soy Lecithin and Propylene Glycol), microcrystalline cellulose and lactose monohydrate. To select the most suitable formulations for extrusion and spheronization, an experimental design of experiences was adopted. The screening amongst formulations (13 different blends) was carried out preparing pellets and evaluating extrusion profiles and quality of the spheronized extrudates. The pellets were characterized for sizing and shape, density and hardness. Although more than one type of pellets demonstrated adequate morphological and technological characteristics, pellets having best properties were selected for further biopharmaceutical investigations, including in vitro dissolution, determination of released droplet size and in vivo trials on rats to study serum and lymph levels after oral administration of the pellets. These preliminary technological and pharmacokinetic data demonstrated that extrusion/spheronization is a viable technology to produce selfemulsifying pellets of good quality and able to improve in vivo oral bioavailability of main components of a phytotherapic extract, also enhancing its lymphatic absorption. The authors thank Indena for the kind gift of Sylibum Marianum dry extract.

The Enhancing Effect of Sodium Glycocholate and Sodium Salicylate on Rats Gastro-intestinal Permeability to Insulin

There have been numerous efforts to formulate insulin into an oral dosage form. The major problems involved with the oral administration of insulin are acidic and enzymatic decomposition by the gastric medium, and poor absorption in the small intestine due to its macromolecular structure. This study attempted to test the enhancing ability of two absorption enhancers, sodium glycocholate (Na-G...

In vivo Evaluation of Self Emulsifying Drug Delivery System for Oral Delivery of Nevirapine

Nevirapine is a highly lipophilic and water insoluble non-nucleoside reverse transcriptase inhibitor used for the treatment of HIV-1 infection. Lymphoid tissue constitutes the major reservoir of HIV virus and infected cells in HIV-infected patients. Self-emulsifying drug delivery system, using long chain triglycerides, is a popular carrier of drugs due to their ability to transport lipophilic d...

Self-Emulsifying Granules and Pellets: Composition and Formation Mechanisms for Instant or Controlled Release

Many articles have been published in the last two decades demonstrating improvement in the dissolution and absorption of low solubility drugs when formulated into self-emulsifying drug delivery systems (SEDDS). Several such pharmaceutical products have appeared in the market for medium dose (Neoral® for Cyclsoprin A, Kaletra® for Lopinavir and Ritonavir), or low dose medications (Rocaltrol® for...
